چکیده

Polymeric paramagnetic ion chelate molecules were synthesized and covalently linked to monoclonal antibodies. The labeled antibodies retained their antigen binding capacity in vitro while carrying up to an average of 50 paramagnetic ion chelates, enabling specifically bound antibody concentrations less than 2.0 microM to significantly reduce proton longitudinal relaxation times.

Nuclear magnetic resonance and gamma camera tumor imaging using gadolinium-labeled monoclonal antibodies.

Chelate-derivatized monoclonal antibody labeled with paramagnetic gadolinium-3+ ion has been evaluated as a tumor-specific contrast-enhancing agent in nuclear magnetic resonance imaging in the Rauscher murine erythroleukemia system. With 10(-7) M concentrations of Gd3+ delivered to the tumor target, a small but reproducible difference in proton relaxation times (T1S) was observed in excised tum...

Role of chelates in treatment of cancer.

Chelates are used in cancer as cytotoxic agent, as radioactive agent in imaging studies and in radioimmunotherapy. Various chelates based on ruthenium, copper, zinc organocobalt, gold, platinum, palladium, cobalt, nickel and iron are reported as cytotoxic agent. Monoclonal antibodies labeled with radioactive metals such as yttrium-90, indium-111 and iodine-131 are used in radioimmunotherapy. Th...

Introduction of new derivatives of Biotin and DTPA for labeling of antibodies with ¹¹¹In to detect malignant tumors [Persian]

Radiolabeled monoclonal antibodies, have created new innovations in diagnosis, research and therapy of disease in last 2 decades. One of the serious limitation of applications of radiolabeled antibodies in vivo is relatively low target to background activity. Various strategies have been proposed to solve this problem including pre-targeting methods that was suggested in 1989. Regarding i...

Development and characterization of anti-renal cell carcinoma x antichelate bispecific monoclonal antibodies for two-phase targeting of renal cell carcinoma.

To test a two-step approach for radioimmunotargeting of renal cell cancer, quadroma cells secreting antichelate x anti-renal cell carcinoma bispecific antibodies were obtained by somatic cell fusion. Five monoclonal antibodies against the chelate 1,4,7-triazaheptane-N,N',N"-pentaacetic acid (DTPA) were produced and characterized. Competitive binding assays indicated that the anti-DTPA antibodie...
